Meineke is best known for his time as a power forward and center for the Fort Wayne Pistons, a franchise that is now known as the Detroit Pistons. He played college basketball at Dayton University, where he was a standout player, leading the team to an NIT championship in 1951.

After his college career at Dayton, Meineke was selected with the seventh overall pick in the 1952 NBA Draft by the Fort Wayne Pistons. Standing tall at 6'7" and weighing 210 pounds, he quickly established himself as a rugged and versatile player. He brought considerable rebounding skills and a tenacious presence on both ends of the court to the Pistons.

During his rookie season, Meineke made an immediate impact, averaging 10.7 points and 9.7 rebounds per game. He was recognized for his strong performance by being awarded the NBA Rookie of the Year for the 1952-1953 season, becoming the first ever recipient of the prestigious award. Meineke continued to play for the Pistons for three more seasons, displaying consistency and dedication to his craft. In the 1955-1956 season, he had the opportunity to showcase his skills on the national stage when the Pistons made it to the NBA Finals. Although they ultimately lost to the Philadelphia Warriors, Meineke's contributions were instrumental in their journey to the championship series.

Following his stint with the Pistons, Meineke played for the Cincinnati Royals (which later became the Sacramento Kings). He retired from professional basketball in 1958 after six seasons. Despite his relatively short NBA career, Don Meineke's impact as an outstanding power forward and relentless rebounder earned him a place in basketball history.
